Description

It is elaborated one-stage method of preparation of ethyl ether of 2,6-dimethyl-4-piperidinomethylnicotinic acid with 70% yield by interaction of 4,5-dichloro-3-penten-2-one with ethyl ether of β-aminocrotonic acid with the subsequent treatment of prepared intermediate by piperidine.

Innovative Aspect and Main Advantages

Simplification of process due to exception of purification and isolation of intermediate ethyl ether of 2,6-dimethyl-4- chlormethylnicotinic acid. An introduction of ethyl ether of 2,6-dimethyl-4-piperidinomethylnicotinic acid into outbred white mice intraperitoneally in 10mg/kg dose shows a neurotropic activity analogous to triphtazine widely using in medical practice and is referred to practically low toxic compounds LD50=260 (200-325) mg/kg and an acute toxicity of analog on action- triphtazine LD50=230 (170-315) mg/kg.

Areas of Application

It can be applied in medicine as neurotropic means and also for preparation of amide and salts of 4-piperidinomethylnicotinic acid.
