Transdermal delivery system containing rimantadine for influenza prevention and treatment

Description

Transdermal delivery of drugs is a novel effective method of pharmacotherapy. The donor company propose usage of this method of drug administration for prevention and treatment of influenza. Rates of morbidity and mortality from influenza-related complications can increase dramatically during influenza pandemics. Rimantadine is licensed in many countries as antiinfluenza drug. However, rimantadine can cause CNS and gastrointestinal adverse reactions when administered orally. Therefore we have elaborated transdermal rimantadine-containing delivery system and tested its efficacy on model of experimental influenza in mice. Plasters containing rimantadine were applied on the shaven backs of mice (Pic. 1) and were retained there. The results of our research had shown high antiinfluenza efficacy of elaborated by us transdermal rimantadine-containing delivery system. Comparative investigation have shown that transdermal application of rimantadine is more effective than oral usage of this drug (Fig.2).

Innovative Aspect and Main Advantages

Transdermal rimantadine-containing delivery system for influenza prevention and therapy is created fo the first time.

The main advantages of transdermal drug delivery are:

  • prolongs the drug’s action;
  • decrease of the drug’s effective dose;
  • absence of the drug’s concentration hopping and maintaining of its concentration in the therapeutic range;
  • it doesn’t hurt the patient;
  • transdermal therapeutic systems (TTS) removing from the skin stops drug penetration to the organism in cause of adverse reactions development;
  • fist-pass effect in the liver is reduced;
  • in TTS can be used drugs which having the high activity but irritate the gastrointestinal tract or have little halflife time;
  • reducing of the adverse reactions.
  • usage of TTS is especially handly in babies.

Areas of Application

This developed transdermal delivery system containing antiviral agent can be used for prevention and therapy of influenza. Similar systems containing other antiviral agents may be elaborated for prevention and therapy of influenza and other actual infections.

Stage of Development

Technology of transdermal rimantadine-containing delivery system is elaborated and tested in animal models and available for demonstration. It is in process of take out of patent (priority date is 28.04.2004).

We look forward on international collaboration for clinical trials and for production of transdermal rimantadine-containing delivery system for influenza prevention and therapy.
